A seasonal theme about him working with one of the Dooms would have large stakes.
While it would be a long shot, Nova would also be an intriguing option.
Though the Sinister Six has had numerous members, keeping it to the most frequent members would be best.

Octavius stands to be a menacing Vanguard whose arms could implement new mechanics.
In this regard,Doc Ock and either Electro or Vulture would be the best.

She is an interesting option as she fits well as either a Vanguard or a Duelist.

For the Skrulls, the best option would be to create a Super Skrull.
The easiest characters to add would be Baron Zemo, Red Skull, or Crossbones.
Knull would be an obvious roster addition, with the flexibility to slot in as a Vanguard or Duelist.
